The mitochondrion that speaks
School textbooks leave the mitochondrion as «the cell’s power plant». True, but incomplete.
The mitochondrion has DNA of its own — small, circular, inherited from ancient bacteria. It was long thought to encode only a handful of respiratory-chain proteins. It turned out otherwise: inside the 12S ribosomal RNA gene hides a short frame encoding MOTS-c.
What is unusual here. Normally the nucleus commands the mitochondria. Here it is the reverse: the mitochondrion reports its state to the nucleus, and the nucleus rebuilds the cell’s work. This is called retrograde signalling.
How it works — step by step
1. Exertion
Muscle works, energy runs short, the mitochondrion comes under pressure.
2. Production
The mitochondrion makes MOTS-c and releases it into the cell and the bloodstream.
3. Interference with the folate cycle
The peptide slows the part of metabolism where DNA building blocks are assembled. A compound called AICAR accumulates.
4. AMPK switches on
AICAR activates AMPK — the internal gauge that fires when energy is short.
5. Metabolism shifts

6. A signal to the nucleus
Part of the peptide travels into the nucleus and switches on stress-adaptation genes.
